Glycosyl Residue Composition Analysis of Recombinant and Native Glycoproteins
Purpose:
Quantitative determination of glycosyl residue and glycosyl-linkage composition analysis in glycoproteins.
Methods Used:
Glycosyl residue composition of glycoproteins is usually done by HPAEC because it has the capacity to detect monosaccharide from a very limited amount of sample. In addition, HPAEC is the method of choice for the determination of sialic acids. The monosaccharide present in glycoproteins can be analyzed also by GC-MS; however, the sample submitted should provide 2 to 8 µg per sugar residue (at least 500 µg) to obtain a reliable composition data.
